Fluorescence Lifetime Imaging Microscopy (FLIM) is an advanced imaging technique that measures the decay time of a fluorophore’s excited state, also called Fluorescence Lifetime. This lifetime is sensitive to the local environment, such as pH, ion concentration, or protein interactions, making FLIM highly useful for studying cellular processes, metabolic state, protein-protein interactions, bindings, etc. Unlike intensity-based fluorescence imaging, FLIM is less affected by variations in fluorophore concentration, light scattering, or photobleaching, but most importantly exploits auto-fluorescence signals. Altogether, these features make FLIM a valid approach for many applications, providing a new layer of information on intensity-based imaging.
